醫(yī)學論文范文:ICAM1在實驗性自身免疫性腦脊髓炎發(fā)病機制中的作用
【摘要】 目的 檢測實驗性自身免疫性腦脊髓炎(EAE)大鼠血腦屏障中血管內皮細胞細胞間粘附分子1(ICAM1)的表達,并分析其表達與EAE發(fā)病的關系。方法 取EAE組和對照組大鼠的腦組織,石蠟切片,免疫組化染色,圖像掃描系統(tǒng)觀測,圖像分析軟件進行灰度分析。結果 EAE組大鼠腦血管內皮細胞上ICAM1的表達明顯深于對照組。實驗組的灰度值為141.47±6.88,而對照組為166.24±7.24,兩組差異非常顯著(P<0.01)。結論 大鼠EAE時腦血管內皮細胞ICAM1的表達上調,提示ICAM1是與EAE發(fā)生相關的重要粘附分子。

【Abstract】 Objective To investigate the relationship between the expression of ICAM1 in the bloodbrain barrier and the experimental autoimmune encephalomyelitis(EAE). Methods Brain of EAE as well as control animals were taken and embedded in paraffin.And the sections were stained by immunohistochemical method. Then the sections were observed by Nikon image scanning system and measured the gray value by IPP image analysis software. Results The expression of ICAM1 in the epithelial cell of endangium of EAE rat's brain was higher than that in control. Conclusion The increase of the expression of ICAM1 in the epithelial cell of endangium of EAE rat's brain indicated that ICAM1 was a necessary adhesion molecule in the developmental process of EAE醫(yī).學.全.在.線zxtf.net.cn.

多發(fā)性硬化癥(multiple sclerosis,MS)可能是一種與遺傳、病毒感染、地理環(huán)境等諸多因素有關的自身免疫性疾。1]。實驗性自身免疫性腦脊髓炎(experimental autoimmune encephalomyelitis,EAE)與MS在臨床、生化、免疫及病理等諸多方面具有相同的特征,因此成為深入研究MS的理想動物模型。在免疫學的研究中,粘附分子在EAE發(fā)病中的作用越來越受到關注。那么,血管內皮細胞細胞間粘附分子1(intercellular adhesion molecular1,ICAM1)在血腦屏障中的表達是否與EAE的發(fā)生有關?本研究采用免疫組化法檢測了ICAM1在血腦屏障中的表達,以探討其與EAE發(fā)病的關系。
1 材料與方法
1.1 材料
1.1.1 實驗動物及分組 雌性Wistar大鼠50只,8~10周齡,體重(192±10)g,隨機分為實驗組(免疫抗原組)和對照組(完全弗氏佐劑/百日咳桿菌組)。實驗組注射牛髓鞘堿性蛋白(myelin basic protein,MBP)+完全弗氏佐劑(Freund’s adjuvant complete,CFA)+百日咳桿菌;對照組注射生理鹽水+CFA+百日咳桿菌,每組各25只。
1.1.2 試劑 ICAM1羊抗鼠IgG多抗(Santa Cruz)、SP通用型試劑盒(ZYMED)。
