醫(yī)學(xué)論文范文:大蒜素對人宮頸癌Hela細(xì)胞增殖影響及其作用機(jī)制
【摘要】 目的 探討大蒜素對體外培養(yǎng)宮頸癌Hela細(xì)胞增殖的影響及其作用機(jī)制。方法 采用四甲基偶氮唑藍(lán)(MTT)法測定不同濃度大蒜素對Hela細(xì)胞增殖抑制率;流式細(xì)胞術(shù)測定細(xì)胞周期變化及凋亡相關(guān)蛋白Bcl2與Bax的表達(dá)。結(jié)果 MTT顯示大蒜素能抑制人宮頸癌Hela細(xì)胞的增殖,并具有時間劑量依賴性關(guān)系;流式細(xì)胞術(shù)測定結(jié)果顯示10 μg/ml大蒜素可明顯誘導(dǎo)宮頸癌Hela細(xì)胞凋亡,并將細(xì)胞周期阻滯在G2/M期;可降低癌基因蛋白Bcl表達(dá)(P<0.01),而促進(jìn)Bax表達(dá)(P<0.01)。結(jié)論 大蒜素對人宮頸癌Hela細(xì)胞增殖具有抑制作用,與細(xì)胞周期阻滯和凋亡相關(guān)蛋白表達(dá)有關(guān)。

【Abstract】 Objective To investigate the influence of Diallyl trisulfide on human cervical carcinoma Hela cells proliferation and its action mechanism.Methods The viability of Hela cells after various treatments were determined by MTT method. Flow cytometry was used to analyze the changes of cell cycle and Bcl2,Bax levels.Results The proliferation of human cervical carcinoma Hela cells were inhibited by Diallul trisulfide. The inhibition was associated with dose and time. The cytometry results showed that: (1)When the cells were treated with Diallyl trisulfide at the concentration of 10μg/ml for 12h and 24h,the percentage of G0/G1 phase cells was decreased, however, that of G2/M phase cells was significantly increased, as compared with those in control group.(2)Diallyl trisulfide downregulated Bcl expression,however,which upregulated Bax expression.Conclusion It is suggested that Diallyl trisulfide can inhibit Hela cell proliferation, which is related with the arrest of cell cycles and down regulation of Bcl2/Bax ratio.

大蒜素(Allicin)是大蒜中主要生物活性成分的總稱,是從大蒜鱗莖中分離出的具有多種生物學(xué)活性的化合物,F(xiàn)代醫(yī)學(xué)研究證實大蒜素具有多種生物活性如(抗菌、消炎、提高機(jī)體免疫力和抗腫瘤的作用[1-3])。體外研究顯示,大蒜素可以通過誘導(dǎo)癌細(xì)胞生成活性氧(ROS),上調(diào)Bax和Bak,激活Caspase3等方式促進(jìn)其凋亡[4,5]。本研究以體外培養(yǎng)的人宮頸腺癌Hela細(xì)胞為材料,探討大蒜素對宮頸癌細(xì)胞增殖的影響并研究其作用機(jī)制。
