醫(yī)學(xué)論文范文:CEECL法同時(shí)測(cè)定復(fù)方磷酸可待因中三種成分的方法學(xué)研究
【摘要】 目的 建立毛細(xì)管電泳電化學(xué)發(fā)光法分離并檢測(cè)復(fù)方磷酸可待因中三種成分,磷酸可待因、馬來(lái)酸溴苯那敏和鹽酸麻黃堿含量的新方法。方法 基于堿性介質(zhì)中,復(fù)方磷酸可待因溶液中三種主要成分對(duì)聯(lián)吡啶釕在鉑電極上的電化學(xué)發(fā)光信號(hào)的增敏作用,且增敏強(qiáng)度和藥物濃度成線性關(guān)系,與毛細(xì)管電泳聯(lián)用,建立了毛細(xì)管電泳電化學(xué)發(fā)光法分離并檢測(cè)其含量的新方法。結(jié)果 在優(yōu)化的實(shí)驗(yàn)條件下,確定三種藥物對(duì)照品的線性范圍、線性方程和檢出限。通過(guò)對(duì)濃度為1.0×10-5kg/L的磷酸可待因、馬來(lái)酸溴苯那敏和鹽酸麻黃堿對(duì)照品進(jìn)行12次平行測(cè)定,其ECL發(fā)光強(qiáng)度的RSD值分別為2.89%,3.76%,3.32%。結(jié)論 該方法可用于復(fù)方磷酸可待因口服溶液中三種主要成分的含量測(cè)定,待測(cè)樣品中的基質(zhì)不干擾測(cè)定,方法的回收率分別為100.4%,101.9%,99.8%(n=5)。

Methodology research on determination of three ingredients in compound codeine phosphate solution by CEECL

ABSTRACT: Objective To establish a novel method for simultaneous determination of three ingredients (codeine phosphate, brompheniramine maleate and ephedrine hydrochloride) in compound codeine phosphate solution by using capillary electrophoresis (CE) coupled with electrochemiluminescence (ECL). Methods Based on enhanced ECL intensity of tris (2,2′bypyridine) ruthenium (Ⅱ) with codeine phosphate, brompheniramine maleate and ephedrine hydrochloride, as well as the linear correlation between enhanced sensitivity intensity and drug concentration, platinum disk (300μm in diameter) was used as a working electrode. Parameters affecting separation and detection were optimized. Results Under the optimized conditions, we determined the linear range, regression equation and detection limit for the three ingredients. The relative standard deviation of ECL intensity for twelve consecutive injections of 1.0×10-5kg/L standard sample of three kinds of drug was 2.89% for codeine phosphate, 3.76% for brompheniramine maleate and 3.32% for ephedrine hydrochloride, respectively. Satisfactory results were obtained without interference from sample matrixes. Conclusion The method was successfully applied to the determination of three ingredients in oral solution of compound codeine phosphate, with the recovery rate of 100.4% for codeine phosphate, 101.9% for brompheniramine maleate and 99.8% for ephedrine hydrochloride醫(yī).學(xué)全.在.線網(wǎng)站zxtf.net.cn.

復(fù)方磷酸可待因口服溶液具有鎮(zhèn)咳祛痰,抗過(guò)敏,治療傷風(fēng),流感之功效。其中主要成分為磷酸可待因、鹽酸麻黃堿、馬來(lái)酸溴苯那敏、愈創(chuàng)木酚甘油醚和輔料,成分較為復(fù)雜,在一般檢測(cè)方法中相互干擾和影響。文獻(xiàn)中已報(bào)道的磷酸可待因的含量測(cè)定方法有非水滴定法,酸堿滴定法[1];鹽酸麻黃堿的含量測(cè)定方法有非水滴定法,分光光度法[1];馬來(lái)酸溴苯那敏的含量測(cè)定方法有非水滴定法[1],分光光度法[12],高效液相色譜法[34],電化學(xué)發(fā)光法[5]。這些方法都有其不足之處,如混合樣品中各成分很難同時(shí)檢測(cè)。本文依據(jù)三種待測(cè)藥物磷酸可待因、鹽酸麻黃堿和馬來(lái)酸溴苯那敏的結(jié)構(gòu)(圖1)中的胺基與電化學(xué)發(fā)光
